I. Background | |
活化 B 細(xì)胞的核因子 κ 輕鏈增強(qiáng)子( NF-κB ) 是一個(gè)轉(zhuǎn)錄因子蛋白復(fù)合物家族,可控制 DNA 轉(zhuǎn)錄、細(xì)胞因子產(chǎn)生和細(xì)胞存活。NF-κB 存在于幾乎所有動(dòng)物細(xì)胞類(lèi)型中,并參與細(xì) 胞對(duì)壓力、細(xì)胞因子、自由基、重金屬、紫外線照射、氧化 LDL 以及細(xì)菌或病毒抗原等刺激的反應(yīng)。 NF-κB 的激活涉及兩條主要信號(hào)通路,即經(jīng)典通路和非經(jīng)典(或替代)通路,盡管它 們的信號(hào)機(jī)制不同,但都對(duì)調(diào)節(jié)免疫和炎癥反應(yīng)十分重要。經(jīng)典 NF-κB 激活的主要機(jī)制是通過(guò)多亞基 IκB 激酶 (IKK) 復(fù)合物對(duì) IκBα 的位點(diǎn)特異性磷酸化引發(fā)的誘導(dǎo)性降解。與經(jīng)典 NF-κB 通路相反,非經(jīng)典 NF-κB 通路選擇性地對(duì)一組特定的刺激作出反應(yīng),包括 TNFR 超 家族成員子集的配體,例如 LTβR、BAFFR、CD40 和 RANK。 |

NFκB-Luc HEK293 報(bào)告基因細(xì)胞,是由 NFκB 調(diào)控并表達(dá) Luc 熒光素酶 報(bào)告基因的 HEK293細(xì)胞。PMA是 DAG(二酯酰甘油)的類(lèi)似物,可自由的通過(guò)細(xì)胞膜, 直接作用于細(xì)胞活化信號(hào)轉(zhuǎn)導(dǎo)通路上的 PKC,原理見(jiàn)下圖所示。 Figure 1:NFκB-Luc HEK293 細(xì)胞模型原理圖 |
